The actual B.C. of a bullet does vary with atmospheric conditions, and with velocity.
If you want to determine an actual B.C. , under a given set of conditions, shoot your load at various points out to 500 yards or so, and using the muzzle velocity from your chronograph, plug values for B.C. into a trajectory calculation, such as one of the JBM versions, and vary the B.C. value, until the calculated trajectory matches your actual trajectory.
I personally don't worry about the number itself, since I don't depend on trajectory calculations. I simply shoot the load at various distances so that I know where the point of impact actually is, not where a calculation determines that the point of impact should be.
